Scientific Disclaimer

Please read this before using calculator results in real experiments.

Results are theoretical estimates

All values produced by the Primer Tm Calculator — melting temperatures, GC content, annealing temperature estimates, and quality indicators — are theoretical estimates generated by mathematical models. They may differ from results obtained with commercial calculators, different parameter sets, or your actual experimental conditions.

Real PCR depends on real conditions

Actual PCR performance depends on many factors the calculator does not model, including your template sequence and complexity, polymerase choice, buffer composition, pH, additives such as DMSO or betaine, cycling parameters, and sample quality. A favorable calculation never guarantees successful amplification, and an unfavorable one does not prove a primer cannot work.

Validate experimentally

Always validate primers and reaction conditions experimentally — for example with gradient PCR, appropriate controls, and specificity checks — before relying on them for important work. This tool supports planning and optimization; it does not replace laboratory optimization or professional judgment.

Not medical advice

Nothing on this site constitutes medical advice, diagnostic guidance, or a recommendation for clinical use. The calculator is an educational tool for research and learning contexts only.

No guaranteed outcomes

We make no guarantees about experimental outcomes, and we are not responsible for failed experiments, wasted reagents, or decisions made on the basis of calculator output. Use the results thoughtfully and at your own discretion.
